The sentence underlines that the book is mine and not yours. "This book is mine." If you want to underline something, especially in written form, put the word at the end of the sentence. If you speak, you can put emphasis with intonation.
and (2) is more neutral in emphasis: stresses that it's my book and not yours? (I'm sure intonation can change the emphasis as well, though.
It's just a neutral phrase. Without any context I can't say what it stresses. "This is my book" - what is stressed here? Use intonation if you want to underline something.
